Background of Lcn2
Lipocalin-2 (LCN2), also known as NGAL, is a protein associated with neutrophil gelatinase.1 The LCN2 gene is located at 9q34 and contains 7 exons.2 The 25-kD LCN2 protein is believed to bind small lipophilic substances such as bacteria-derived lipopolysaccharide (LPS) and formylpeptides and may function as a modulator of inflammation. NGAL tightly binds bacterial catecholate-type ferric siderophores through a cyclically permuted, hybrid electrostatic/cation-pi interaction and is a potent bacteriostatic agent in iron-limiting conditions.3 The primary LCN2 transcript is 3,696 nucleotides long, and the processed transcript is 809 nucleotides long.4 LCN2 expression in adult bone marrow, uterus, prostate, salivary gland, stomach, appendix, colon, trachea, and lung, and in fetal spleen and lung. The standard product used in this kit is recombinant rat NGAL, consisting of 177 amino acids with the molecular mass of 21kDa.

10 Customer Q&As for Rat Lipocalin-2/NGAL ELISA Kit PicoKine®
Question
Q: Are Boster Bio recombinant proteins and antibodies sterile?
Verified Customer
Verified customer
Asked: 2020-04-22
Answer
A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

Question
Can EK0855 be used for stool supernatant samples?
Verified customer
Asked: 2020-01-17
Answer
Our lab hasn't worked on stool supernatant samples with the Rat Lipocalin-2/NGAL ELISA Kit PicoKine™ (EK0855). We suggest to run a pilot test. In theory, the kit is able to detect the target protein if the expression level of NGAL in the stool supernatant samples falls in the assay range of EK0855.

Question
Is there supposed to be a washstep in between step 5 and step 6 of the ELISA protocols in the datasheet?
Verified customer
Asked: 2019-04-09
Answer
There is no wash step in between step 5 and step 6 of the ELISA protocols in the datasheet.

Question
Q: if the enzyme conjugated Lipocalin-2 antibodies are mixed with the substrate, will that change the substrate into the enzymatic reaction product? Or the enzyme function is only activated when the antibody is attached with the Lipocalin-2 antigen?
Verified Customer
Verified customer
Asked: 2017-07-17
Answer
A: The enzyme is always active. Avoid contaminating the substrate with enzyme prior to the incubation otherwise it compromises the assay with false positive signal.

Question
I am preparing to use your Rat Lcn2 ELISA cat#EK0855. In reading the procedure I noticed there is not a wash after step 5 and before step 6. This is after incubating samples/standard and before addition of the 1xbiotinylated Anti-Rat Lcn2 antibody. Is this correct?

Question
Q: how to thaw whole blood sample for Lipocalin-2 ELISA after freezing?
Verified Customer
Verified customer
Asked: 2017-06-14
Answer
A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test Lipocalin-2 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

Question
Q: how many samples can be assayed in a Picokine® ELISA Kit?
Verified Customer
Verified customer
Asked: 2016-04-12
Answer
A: The Picokine® ELISA Kits will generally run a 7-point standard curve, non-specific binding wells, and 39 samples in duplicate. this may might differ by kit so please refer to each datasheet for details.

Question
The customer would like to know if (#LB001-02 from Welgene) can be used as washing buffer for EK0855 kit. The composition is 200mg/L KCl, 200mg/L KH2PO4, 8000mg/L NaCl, 2160mg/L Na2HPO4
Verified Customer

Asked: 2014-12-26
Answer
The recipe of our wash buffer is different from that of LB002-01. We haven't used the same buffer as LB002-01. And we do not suggest the customer use it. Please use the wash buffer included in the kit. Or prepare wash buffer as the method described below. Washing buffer (neutral PBS or TBS). ? Preparation of 0.01M TBS: Add 1.2g Tris, 8.5g Nacl; 450?l of purified acetic acid or 700?l of concentrated hydrochloric acid to 1000ml H2O and adjust pH to 7.2-7.6. Finally, adjust the total volume to 1L. ? Preparation of 0.01M PBS: Add 8.5g sodium chloride, 1.4g Na2HPO4 and 0.2g NaH2PO4 to 1000ml distilled water and adjust pH to 7.2-7.6. Finally, adjust the total volume to 1L.

Question
Q: is it okay to use citrate plasma as samples in Rat Lipocalin-2 Picokine® ELISA Kit (Catalog # EK0855)?
K. Scott
Verified customer
Asked: 2014-05-18
Answer
A: Chelating agents such as EDTA, Heparin and Citrate can bind metal ions from the functional domain of Lipocalin-2 causing disruption of its protein structure. Lipocalin-2 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes a must for detection and block the antigen antibody reaction. We have tested the Lipocalin-2 ELISA, treating samples with different anticoagulants and decided that heparin can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.
