Background of Angiopoietin-like protein 8/Betatrophin
ANGPTL8 (also known as lipasin, originally Betatrophin) is a protein that in humans is encoded by the C19orf80 gene. The gene for betatrophin lies on mouse chromosome 9 (gene symbol: Gm6484) and on human chromosome 19 (gene symbol: C19orf80). The encoded 22 kDa protein contains an N-terminal secretion signal and two coiled-coil domains and is a member of the angiopoietin-like (ANGPTL) protein family. However, in contrast to other ANGPTL proteins, betatrophin lacks the C-terminal fibrinogen-like domain, and therefore it is an atypical member of the ANGPTL family. It shares with Angptl4 and Angptl3 the ability to inhibit the enzyme Lipoprotein lipase (LPL), and its hepatic overexpression causes elevation of circulating Triglyceride levels in mice. In mice betatrophin is secreted by the liver. It was hoped that betatrophin or its homolog in humans may provide an effective treatment for type 2 diabetes and perhaps even type I diabetes.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human HepG2 whole Cell lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-C19orf80 antigen affinity purified polyclonal antibody (Catalog # A02471) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for C19orf80 at approximately 22KD. The expected band size for C19orf80 is at 22KD.

You can run a BLAST between primate and the immunogen sequence of anti-C19orf80/ANGPTL8 antibody A02471 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated primate samples, this use of the antibody is not covered by our guarantee.
