Background of cIAP-2/HIAP-1
Baculoviral IAP repeat-containing protein 3 (also known as cIAP2) is a protein that in humans is encoded by the BIRC3 gene. cIAP2 is a member of the IAP family of proteins that inhibit apoptosis by binding to tumor necrosis factor receptor-associated factors TRAF1 and TRAF2, probably by interfering with activation of ICE-like proteases. The encoded protein inhibits apoptosis induced by serum deprivation but does not affect apoptosis resulting from exposure to menadione, a potent inducer of free radicals. It contains 3 baculovirus IAP repeats and a ring finger domain. Transcript variants encoding the same isoform have been identified.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: HEPG2 Whole Cell Lysate at 40ug,
Lane 2: HELA Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-cIAP2 antigen affinity purified polyclonal antibody (Catalog # PB9527) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for cIAP2 at approximately 68 kDa. The expected band size for cIAP2 is at 68 kDa.

We are currently using anti-cIAP2/BIRC3 antibody PB9527 for human tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on dog tissues as well?

The anti-cIAP2/BIRC3 antibody (PB9527) has not been tested for cross reactivity specifically with dog tissues, but there is a good chance of cross reactivity.
