Background of CXCL9/MIG
MIG, also known as CXCL9, is a T-cell chemoattractant inducible by gamma interferon that is a member of the CXC chemokine family of cytokines. This gene is mapped to 4q21. It is noted that, although the best-described activities of the chemokines are as chemotactic factors, chemokines also have an effect on T-cell activation, angiogenesis, and HIV infection. While most CXC chemokines are chemotactic for neutrophils, MIG and INP10 are unusual and similar in being CXC chemokines that are chemotactic for lymphocytes and inactive in neutrophils.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Human Placenta Tissue Lysate at 50ug,
Lane 2: A431 Whole Cell Lysate at 40ug,
Lane 3: HELA Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CXCL9 antigen affinity purified polyclonal antibody (Catalog # PB9694) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CXCL9 at approximately 19 kDa. The expected band size for CXCL9 is at 14 kDa.

We are currently using anti-MIG/CXCL9 antibody PB9694 for human tissue, and we are satisfied with the ELISA results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on feline tissues as well?

The anti-MIG/CXCL9 antibody (PB9694) has not been validated for cross reactivity specifically with feline tissues, but there is a good chance of cross reactivity.
