IHC analysis of Cytokeratin 6B/KRT6B using anti-Cytokeratin 6B/KRT6B antibody (A07579-1).
Cytokeratin 6B/KRT6B was detected in a paraffin-embedded section of human skin tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1:50 rabbit anti-Cytokeratin 6B/KRT6B Antibody (A07579-1) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB as the chromogen.

Background of KRT6B
Keratins are a large family of proteins that form the intermediate filament cytoskeleton of epithelial cells, which are classified into two major sequence types. Type I keratins are a group of acidic intermediate filament proteins, including K9-K23, and the hair keratins Ha1-Ha8. Type II keratins are the basic or neutral courterparts to the acidic type I keratins, including K1-K8, and the hair keratins, Hb1-Hb6. Keratin 6 is a type II keratin. It is used as a marker for epidermal hyperproliferation and differentiation. Three keratin 6 isoforms were found, ketatin 6A,6B, and 6C. They share more than 99% identical DNA sequence.
