Background of FBXO43
Fbp5B (also called F-Box protein 43 or Fbx043) is a relatively new member of the F-box protein family that is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of the ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbxs class.

Assay Dilutions Recommendation

This affinity-purified antibody has been tested for use in ELISA and western blot. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 105 kDa in size corresponding to Fbp5B protein by western blotting in the appropriate cell lysate or extract. Predicted molecular weight is 51 kDa.

Western blot using Boster's affinity purified anti-Fbp5B antibody. Panels A and B show two identical membranes containing marker, in vitro translated Fbp5b (Fbp5b ivt), and a whole cell extract from 293T cells loaded in duplicate). Panel A shows antibody reactivity. Panel B shows antibody reactivity after first pre-incubating the antibody with the immunizing peptide. This pre-incubation greatly diminishes specific band recognition. Peptide inhibition completely removes the in vitro translated band and greatly reduces the endogenous band from 293T cells. We are currently using anti-Fbp5B antibody A12000 for rat tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on pig tissues as well?
