3. Flow Cytometry analysis of HEPG2 cells using anti-Fetuin A antibody (PB9568).
Overlay histogram showing HEPG2 cells stained with PB9568 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-Fetuin A Antibody (PB9568,1μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1μg/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.

Background of AHSG
Alpha-2-HS-glycoprotein (AHSG), also known as fetuin-A, is a protein that in humans is encoded by the AHSG gene. Fetuin-A belongs to the fetuin class of plasma binding proteins and is more abundant in fetal than adult blood. Its gene is mapped to 3q27. The protein is commonly present in the cortical plate of the immature cerebral cortex and bone marrow hemopoietic matrix. It is involved in several functions, such as endocytosis, brain development and the formation of bone tissue.
