Background of GAB1
GRB2-associated-binding protein 1 is a protein that in humans is encoded by the GAB1 gene. It is mapped to 4q31.21. The protein encoded by this gene is a member of the IRS1-like multisubstrate docking protein family. It is an important mediator of branching tubulogenesis and plays a central role in cellular growth response, transformation and apoptosis. Two transcript variants encoding different isoforms have been found for this gene.

Western blot analysis of GAB1 using anti-GAB1 antibody (A01989-1).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
lane 1: rat testis tissue lysates,
lane 2: K562 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GAB1 antigen affinity purified polyclonal antibody (Catalog # A01989-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for GAB1 at approximately 77KD. The expected band size for GAB1 is at 77KD.

We are currently using anti-GAB1 antibody A01989-1 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on dog tissues as well?

The anti-GAB1 antibody (A01989-1) has not been validated for cross reactivity specifically with dog tissues, but there is a good chance of cross reactivity.
