Flow Cytometry analysis of U937 cells using anti-SLC2A9 antibody (A02245).
Overlay histogram showing U937 cells stained with A02245 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-SLC2A9 Antibody (A02245, 1μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

Background of SLC2A9
Solute carrier family 2, facilitated glucose transporter member 9, also known as SLC2A9, is a protein that in humans is encoded by the SLC2A9 gene. This gene encodes a member of the SLC2A facilitative glucose transporter family. Members of this family play a significant role in maintaining glucose homeostasis. This gene is mapped to 4p16.1. The encoded protein may play a role in the development and survival of chondrocytes in cartilage matrices. This gene transports urate and fructose. It may have a role in the urate reabsorption by proximal tubules. This gene also transports glucose at low rate.
