Background of GPR116

Anti-CAND2 antibody is ideal for western blotting, ELISA and IHC. CAND2 is also known as TIP120B, and TATA-binding protein-interacting protein 120B. While both CAND1 (TIP120A) and CAND2 (TIP120B) are TATA-binding proteins and form complexes with various nuclear proteins involved in the control of eukaryotic gene transcription, CAND2 (TIP120B) is expressed specifically in muscle and heart tissue. This is contrary to the ubiquitous expression of CAND1 (TIP120A). TIP120 homologs exist in various higher eukaryotes including D. melanogaster, C. elegans, and A. thaliana. TIP120B is 60% identical in amino acid sequence to TIP120A.

You can run a BLAST between canine and the immunogen sequence of anti-GPR116 antibody A10539 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated canine samples, this use of the antibody is not covered by our guarantee. Does A10539 anti-GPR116 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer

As indicated on the product datasheet, A10539 anti-GPR116 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
