Background of CDC73/HRPT2
Cell division cycle 73, also known as HRPT2, is a protein which in humans is encoded by the CDC73 gene. This gene encodes a tumor suppressor that is involved in transcriptional and post-transcriptional control pathways. The protein is a component of the the PAF protein complex, which associates with the RNA polymerase II subunit POLR2A and with a histone methyltransferase complex. And this protein appears to facilitate the association of 3' mRNA processing factors with actively-transcribed chromatin. Mutations in this gene have been linked to hyperparathyroidism-jaw tumor syndrome, familial isolated hyperparathyroidism, and parathyroid carcinoma.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Human Placenta Tissue Lysate at 50ug,
Lane 2: HEPG2 Whole Cell Lysate at 40ug,
Lane 3: 293T Whole Cell Lysate at 40ug,
Lane 4: HELA Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-HRPT2 antigen affinity purified polyclonal antibody (Catalog # PB9532) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for HRPT2 at approximately 61 kDa. The expected band size for HRPT2 is at 61 kDa.

Question
We are currently using anti-HRPT2/CDC73 antibody PB9532 for human tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on zebrafish tissues as well?

The anti-HRPT2/CDC73 antibody (PB9532) has not been tested for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity.
