Storage & Handling

Store at -20˚C for one year from date of receipt. After reconstitution, at 4˚C for one month. It can also be aliquotted and stored frozen at -20˚C for six months. Avoid repeated freeze-thaw cycles.

Background of HTT

The huntingtin gene, also called HTT or HD (Huntington disease) gene, is the IT15 ("interesting transcript 15") gene which codes for a protein called the huntingtin protein. It is mapped to 4p16.3. The protein has no sequence homology with other proteins and is highly expressed in neurons and tests in humans and rodents. HTT upregulates the expression of Brain Derived Neurotrophic Factor (BDNF) at the transcription level, and this gene is required for normal development. The HTT protein is involved in vesicle trafficking as it interacts with HIP1, a clathrin-binding protein, to mediate endocytosis, the absorption of materials into a cell. HTT was also visualized as punctate staining likely to represent nerve endings. What's more, wildtype HTT may function in the nucleus in the assembly of nuclear matrix-bound protein complexes involved with transcriptional repression and RNA processing.

My colleagues were well pleased with the WB result of your anti-Huntingtin/HTT antibody. However we have observed positive staining in cervix carcinoma erythroleukemia myristoylated n-terminal using this antibody. Is that expected? Could you tell me where is HTT supposed to be expressed?

Answer

According to literature, cervix carcinoma erythroleukemia does express HTT. Generally HTT expresses in huntingtin: cytoplasm, huntingtin, myristoylated n-terminal. Regarding which tissues have HTT expression, here are a few articles citing expression in various tissues:

We are currently using anti-Huntingtin/HTT antibody PA2268 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on horse tissues as well?

The anti-Huntingtin/HTT antibody (PA2268) has not been validated for cross reactivity specifically with horse tissues, but there is a good chance of cross reactivity. Based on Uniprot.org, HTT is expressed in cerebellum, retina, brain, brain, caudate nucleus, frontal cortex, muscle retina, platelet, cervix carcinoma, leukemic t-cell, cervix carcinoma erythroleukemia, liver, among other tissues.
