Background of Slc22a1
Solute carrier family 22(organic cation transporter), member 1, also called SLC22A1 or OCT1 is a protein that in humans is encoded by the SLC22A1 gene. This gene is mapped to 6q25.3. Polyspecific organic cation transporters in the liver, kidney, intestine, and other organs are critical for elimination of many endogenous small organic cations as well as a wide array of drugs and environmental toxins. This gene transports the polyamines spermine and spermidine. It also transports pramipexole across the basolateral membrane of the proximal tubular epithelial cells. This gene regulated by various intracellular signaling pathways including inhibition by protein kinase A activation, and endogenously activation by the calmodulin complex, the calmodulin-dependent kinase II and LCK tyrosine kinase.

Western blot analysis of OCT1/Slc22a1 using anti-OCT1/Slc22a1 antibody (A01945-1).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: mouse HEPA1-6 whole cell lysates.
red to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-OCT1/Slc22a1 antigen affinity purified polyclonal antibody (Catalog # A01945-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for OCT1/Slc22a1 at approximately 75 kDa. The expected band size for OCT1/Slc22a1 is at 62 kDa.
