Background of DLG4
DLG4 discs large homolog 4, also known as PSD95 or SAP-90, is a protein that in humans is encoded by the DLG4 gene. It is a member of the membrane-associated guanylate kinase (MAGUK) family. This gene is mapped to 17p13.1. DLG4 can heteromultimerize with another MAGUK protein, DLG2, and is recruited into NMDA receptor and potassium channel clusters. These two MAGUK proteins may interact at postsynaptic sites to form a multimeric scaffold for the clustering of receptors, ion channels, and associated signaling proteins. Overexpression of DLG4 in hippocampal neurons could drive maturation of glutamatergic synapses. DLG4 can orchestrate synaptic development and it has a role in synapse stabilization and plasticity. Ubiquitination of DLG4 through an MDM2-mediated pathway can regulate AMPA receptor surface expression during synaptic plasticity.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Western blot analysis of PSD95 using anti-PSD95 antibody (PB9802).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: Rat Brain Tissue Lysate,
Lane 2: mouse brain Tissue Lysate,
Lane 3: human SHG-44 Whole Cell Lysate,
Lane 4: human U-87MG Whole Cell Lysate
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PSD95 antigen affinity purified polyclonal antibody (Catalog # PB9802) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PSD95 at approximately 100KD. The expected band size for PSD95 is at 80KD.

Flow Cytometry analysis of A431 cells using anti-PSD95 antibody (PB9802).
Overlay histogram showing A431 cells stained with PB9802 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-PSD95 Antibody (PB9802,1μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

Question
We are currently using anti-PSD95/DLG4 antibody PB9802 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on canine tissues as well?

Answer
The anti-PSD95/DLG4 antibody (PB9802) has not been validated for cross reactivity specifically with canine tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in canine you can get your next antibody for free. Please contact me if I can help you with anything.
