Background of PTPN7
PTPN7, also known as haematopoietic PTP (HePTP), belongs to the R7 family of protein tyrosine phosphatases (PTPs) which all contain a conserved 16 amino acid sequence at their N-termini (PMID: 9857190). PTPN7, via negative regulation of MAPK signalling, plays a key role in T-cell and B-cell development and is the only known PTP whose expression in T-lymphocytes is induced by IL-2 (PMID: 15980098). PTPN7 is a 38 kDa non-receptor PTP expressed predominately in leukocytes, spleen and thymus (PMID: 1530918, 24265715, 19636879).

Western blot analysis of PTPN7 using anti-PTPN7 antibody (A07600-1).
Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human THP-1 whole cell lysates,
Lane 2: human HEL whole cell lysates,
Lane 3: human Jurkat whole cell lysates,
Lane 4: human K562 whole cell lysates,
Lane 5: rat thymus tissue lysates,
Lane 6: mouse spleen tissue lysates,
Lane 7: mouse thymus tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PTPN7 antigen affinity purified polyclonal antibody (A07600-1) at 1:1000 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for PTPN7 at approximately 38-40 kDa. The expected band size for PTPN7 is at 41 kDa.
