Background of RGS3

RGS3 (Regulator of G Protein Signaling 3), is a protein that in humans is encoded by the RGS3 gene. Chatterjee et al. (1997) studied RGS3, the largest member of the RGS family then identified. Mouse Pdz-Rgs3 can mediate signaling from the ephrin-B cytoplasmic tail. By radiation hybrid mapping, Chatterjee et al. (1997) assigned the RGS3 gene to chromosome 9q31-q33. Lu et al. (2001) showed that Sdf1, a chemokine with a G protein-coupled receptor, and Bdnf are chemoattractants for cerebellar granule cells, and that Sdf1 chemoattraction is selectively inhibited by soluble ephrin-B receptor. This inhibition could be blocked by a truncated Pdz-Rgs3 protein lacking the RGS domain.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

4 Customer Q&As for Anti-RGS3 Antibody

Question

Does anti-RGS3 antibody PA2050 work on feline WB with brain kidney?

Verified Customer

Verified customer

Asked: 2019-01-18

Answer

Our lab technicians have not validated anti-RGS3 antibody PA2050 on feline. You can run a BLAST between feline and the immunogen sequence of anti-RGS3 antibody PA2050 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated feline samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in feline brain kidney in WB, you can get your next antibody for free.

Question

We are currently using anti-RGS3 antibody PA2050 for rat tissue, and we are satisfied with the IHC results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on feline tissues as well?

Verified Customer

Asked: 2017-05-31

Answer

The anti-RGS3 antibody (PA2050) has not been validated for cross reactivity specifically with feline tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in feline you can get your next antibody for free. Please contact me if I can help you with anything.

Question

Will PA2050 anti-RGS3 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

J. Yang

Verified customer

Asked: 2014-09-30

Answer

As indicated on the product datasheet, PA2050 anti-RGS3 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
