Background of RPL34
Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal protein that is a component of the 60S subunit. The protein belongs to the L34E family of ribosomal proteins. It is located in the cytoplasm. This gene originally was thought to be located at 17q21, but it has been mapped to 4q. Transcript variants derived from alternative splicing, alternative transcription initiation sites, and/or alternative polyadenylation exist; these variants encode the same protein. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome.

Would A08190 anti-RPL34/Ribosomal Protein L34 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
It shows on the product datasheet, A08190 anti-RPL34/Ribosomal Protein L34 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-RPL34/Ribosomal Protein L34 antibody A08190 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on primate tissues as well?

The anti-RPL34/Ribosomal Protein L34 antibody (A08190) has not been tested for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity. Our lab technicians have not tested anti-RPL34/Ribosomal Protein L34 antibody A08190 on primate. You can run a BLAST between primate and the immunogen sequence of anti-RPL34/Ribosomal Protein L34 antibody A08190 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated primate samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in primate liver in WB, you can get your next antibody for free.
