Flow Cytometry analysis of SH-SY5Y cells using anti-SCLY antibody (A09046).
Overlay histogram showing SH-SY5Y cells stained with A09046 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-SCLY Antibody (A09046, 1 μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10 μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1 μg/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.

Use TE buffer pH 9.0 for antigen retrieval; (*) citrate buffer pH 6.0 is an alternative.

Background of SCLY
SCLY(Selenocysteine Lyase), is an enzyme that catalyzes the chemical reaction. This enzyme belongs to the family of lyases, specifically the class of carbon-sulfur lyases. The International Radiation Hybrid Mapping Consortium mapped the human SCLY gene to chromosome 2. Mihara et al.(2000) showed that mouse Scly catalyzed the conversion of L-selenocysteine to L-alanine. Scly activity required pyridoxal 5-prime phosphate, was specific to L-selenocysteine, and showed maximum reactivity at pH 9.0.
