Background of SCTR
Human secretin receptor (gene name SCTR) is a G protein-coupled receptor and belongs to the glucagon-VIP-secretin receptor family. It binds secretin which is the most potent regulator of pancreatic bicarbonate, electrolyte and volume secretion. Secretin and its receptor are suggested to be involved in pancreatic cancer and autism. The SCTR gene is mapped to chromosome 2q14.1 by fluorescence in situ hybridization.

Western blot analysis of SCTR using anti-SCTR antibody (PB10096).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat kidney tissue lysates,
Lane 2: SKOV3 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SCTR antigen affinity purified polyclonal antibody (Catalog # PB10096) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for SCTR at approximately 59 kDa. The expected band size for SCTR is at 50 kDa.

Our lab technicians have not tested anti-SCTR antibody PB10096 on monkey. You can run a BLAST between monkey and the immunogen sequence of anti-SCTR antibody PB10096 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated monkey samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in monkey pancreas in WB, you can get your next antibody for free.

We are currently using anti-SCTR antibody PB10096 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on horse tissues as well?
