Background of SLC5A3

This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbls class and, in addition to an F-box, contains several tandem leucine-rich repeats and is localized in the nucleus. "Winston J.T.,et al.(1999) Curr. Question

We are currently using anti-SMIT antibody A08337 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on zebrafish tissues as well?

Verified Customer

Verified customer

Asked: 2019-12-27

Answer

The anti-SMIT antibody (A08337) has not been tested for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in zebrafish you can get your next antibody for free. Please contact me if I can help you with anything.

Question

Would A08337 anti-SMIT antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2018-02-22

Answer

You can see on the product datasheet, A08337 anti-SMIT antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
