Background of MAP3K7
Mitogen-activated protein kinase kinase kinase 7, also known as TAK1, is an enzyme that in humans is encoded by the MAP3K7 gene. The protein encoded by this gene is a member of the serine/threonine protein kinase family. This kinase mediates the signaling transduction induced by TGF beta and morphogenetic protein (BMP), and controls a variety of cell functions including transcription regulation and apoptosis. In response to IL-1, this protein forms a kinase complex including TRAF6, MAP3K7P1/TAB1 and MAP3K7P2/TAB2; this complex is required for the activation of nuclear factor kappa B. This kinase can also activate MAPK8/JNK, MAP2K4/MKK4, and thus plays a role in the cell response to environmental stresses. Four alternatively spliced transcript variants encoding distinct isoforms have been reported.

Western blot analysis of TAK1/MAP3K7 using anti-TAK1/MAP3K7 antibody (PB10006).
Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human MCF-7 whole cell lysates,
Lane 2: human A431 whole cell lysates,
Lane 3: human A375 whole cell lysates,
Lane 4: human COLO320 whole cell lysates,
Lane 5: rat heart tissue lysates,
Lane 6: rat H9C2 whole cell lysates,
Lane 7: mouse heart tissue lysates,
Lane 8: mouse NIH/3T3 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TAK1/MAP3K7 antigen affinity purified polyclonal antibody (Catalog # PB10006) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for TAK1/MAP3K7 at approximately 75 kDa. The expected band size for TAK1/MAP3K7 is at 67 kDa.

IF analysis of TAK1/MAP3K7 using anti-TAK1/MAP3K7 antibody (PB10006).
TAK1/MAP3K7 was detected in an immunocytochemical section of U2OS cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent (AR0022) for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 5 μg/mL rabbit anti-TAK1/MAP3K7 Antibody (PB10006) overnight at 4°C. DyLight®488 Conjugated Goat Anti-Rabbit IgG (BA1127) was used as secondary antibody at 1:500 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

Flow Cytometry analysis of A375 cells using anti-TAK1/MAP3K7 antibody (PB10006).
Overlay histogram showing A375 cells stained with PB10006 (Blue line). The cells were fixed with 4% paraformaldehyde and blocked with 10% normal goat serum. And then incubated with rabbit anti-TAK1/MAP3K7 Antibody (PB10006, 1 μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10 μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1 μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

We are currently using anti-TAK1/MAP3K7 antibody PB10006 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on monkey tissues as well?

The anti-TAK1/MAP3K7 antibody (PB10006) has not been validated for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity.
