Immunogen
A synthesized peptide derived from human TRIB3 Disrupts insulin signaling by binding directly to Akt kinases and blocking their activation. May bind directly to and mask the 'Thr-308' phosphorylation site in AKT1. Binds to ATF4 and inhibits its transcriptional activation activity. Interacts with the NF-kappa-B transactivator p65 RELA and inhibits its phosphorylation and thus its transcriptional activation activity.
Reactive Species

Western blot analysis of TRIB3 using anti-TRIB3 antibody (M01414).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human K562 whole cell lysates,
Lane 2: human CACO-2 whole cell lysates,
Lane 3: human 293T whole cell lysates,
Lane 4: human HepG2 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TRIB3 antigen affinity purified monoclonal antibody (Catalog # M01414) at 1:1000 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:1000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TRIB3 at approximately 40 kDa. The expected band size for TRIB3 is at 40 kDa.

Question
Will anti-TRIB3 Monoclonal antibody M01414 work on horse IP with liver?
Verified Customer
Verified customer
Asked: 2020-02-03
Answer
Our lab technicians have not tested anti-TRIB3 Monoclonal antibody M01414 on horse. You can run a BLAST between horse and the immunogen sequence of anti-TRIB3 Monoclonal antibody M01414 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated horse samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in horse liver in IP, you can get your next antibody for free.

Question
Would M01414 anti-TRIB3 Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
A. Krishna
Verified customer
Asked: 2018-08-16
Answer
You can see on the product datasheet, M01414 anti-TRIB3 Monoclonal antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
