Background of TNFSF12
Tumor necrosis factor ligand superfamily member 12 also known as TNF-related weak inducer of apoptosis (TWEAK) is a protein that in humans is encoded by the TNFSF12 gene. The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This protein is a ligand for the FN14/TWEAKR receptor. This cytokine has overlapping signaling functions with TNF, but displays a much wider tissue distribution. This cytokine, which exists in both membrane-bound and secreted forms, can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator of angiogenesis. Alternative splicing results in multiple transcript variants.

Western blot analysis of TWEAK using anti-TWEAK antibody (A02009-1).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human MCF-7 cell lysate,
Lane 2: human PANC-1 cell lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TWEAK antigen affinity purified polyclonal antibody (Catalog # A02009-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TWEAK at approximately 27KD. The expected band size for TWEAK is at 27KD.

We are currently using anti-TWEAK/TNFSF12 antibody A02009-1 for human tissue, and we are satisfied with the ELISA results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on canine tissues as well?

The anti-TWEAK/TNFSF12 antibody (A02009-1) has not been tested for cross reactivity specifically with canine tissues, but there is a good chance of cross reactivity.
