Background of CDH5
CDH5 (Cadherin 5), also known as VE-cadherin, is a type of cadherin. It is encoded by the human gene CDH5. Kremmidiotis et al. (1998) mapped the human CDH5 gene to 16q22.1 using somatic cell hybrid panels. Functioning as a classic cadherin by imparting to cells the ability to adhere in a homophilic manner, the protein may play an important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. Therefore it was concluded that VE-cadherin serves the purpose of maintaining newly formed vessels.

Western blot analysis of CDH5 using anti-CDH5 antibody (PA2079).
Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human SK-HEP1 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CDH5 antigen affinity purified polyclonal antibody (PA2079) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054) at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for CDH5 at approximately 120 kDa. The expected band size for CDH5 is at 88 kDa.

IF analysis of CDH5 using anti-CDH5 antibody (PA2079).
CDH5 was detected in an immunocytochemical section of human SK-HEP1 cells. Cells were permeabilized with Triton X-100 for 10 minutes. The cells were blocked with 10% goat serum. And then incubated with 10 μg/mL rabbit anti-CDH5 Antibody (PA2079) overnight at 4°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127), Cy3 conjugated goat anti-mouse IgG (BA1031) was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

You can see on the product datasheet, PA2079 anti-VE Cadherin/CDH5 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-VE Cadherin/CDH5 antibody PA2079 for mouse tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on goat tissues as well?

You can run a BLAST between goat and the immunogen sequence of anti-VE Cadherin/CDH5 antibody PA2079 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated goat samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in goat endothelial cell in WB, you can get your next antibody for free.
