Polyglutamylase that preferentially modifies alpha- tubulin (PubMed:17499049, PubMed:21074048). Mediates tubulin polyglutamylation in cilia (PubMed:22246503).

Involved from the side-chain elongation step of this polyglutamylation reaction rather than from the initiation step (PubMed:17499049, PubMed:21074048). Generates long side-chains (PubMed:20530212).
