What Is the Expected ULBP1 Staining Pattern?

ULBP1 is a GPI-anchored cell-surface protein with no transmembrane segment; UniProt also lists endoplasmic reticulum localization (UniProt Q9BZM6 topology and subcellular location). In tissue IHC, expect cytoplasmic or membranous staining in elongated or late spermatids and Purkinje cells, with staining also reported in lung macrophages (HPA tissue IHC). HPA rates its tissue pattern Enhanced, while noting medium consistency with RNA data and disregarded presumed off-target binding (HPA tissue IHC).

What am I looking at on my slide?
High staining in elongated or late spermatids, with a cytoplasmic or membranous pattern in seminiferous ducts (HPA tissue IHC).This is the clearest supplied tissue reference for a positive IHC result (HPA: High in elongated or late spermatids). Judge staining in the identified cells and compartments, rather than treating uniform staining across the whole duct as the expected pattern (HPA tissue IHC).
Medium cytoplasmic or membranous staining in cerebellar Purkinje cells, or medium staining in lung macrophages (HPA tissue IHC).These are additional reported positive cell populations (HPA tissue IHC). Their lower reported levels make them less straightforward intensity references than late spermatids; compare like cells and regions when reading a section (HPA tissue IHC; general IHC practice).
Strong, predominantly nuclear staining in an otherwise positive region (general IHC practice).A nuclear-dominant pattern conflicts with the reported cytoplasmic and membranous tissue pattern and the listed cell-membrane localization (HPA tissue IHC; UniProt Q9BZM6 subcellular location). Treat it as suspect and assess staining controls before assigning it to ULBP1 (general IHC practice).
Prominent staining in adipocytes or other cells reported as not detected, especially without the expected positive-cell pattern (HPA tissue IHC).HPA reports no detection in adipocytes of adipose tissue or breast and notes presumed off-target binding in its tissue assessment (HPA tissue IHC). Consider cross-reactivity or endogenous detection activity; identify which cells stain and inspect the appropriate detection controls (general IHC practice).
Diffuse stain across many compartments, or no stain in a section containing late spermatids (HPA tissue IHC; general IHC practice).Diffuse staining gives weak evidence of cell-specific localization; absent spermatid staining conflicts with the reported High reference pattern (HPA tissue IHC). Review section quality, primary-antibody and detection controls, and staining conditions before interpreting either result (general IHC practice).
💡Expected ULBP1 appearanceCall positive IHC when elongated or late spermatids show High cytoplasmic or membranous staining, with medium Purkinje-cell staining as another reported pattern; strong nuclear or widespread staining in HPA-negative cell types is suspect (HPA tissue IHC; UniProt Q9BZM6 subcellular location).
How each factor affects the staining
Which cell and tissue are being scored?HPA reports High staining in late spermatids, medium in Purkinje cells and lung macrophages, and no detection in selected adipocytes and glandular cells (HPA tissue IHC). Score the identified cell population; a tissue-wide label can obscure these differences (general IHC practice).
What compartment fits the protein?ULBP1 has a signal peptide, a processed mature chain and a GPI-anchor annotation, with no transmembrane segment (UniProt Q9BZM6 topology and processing). Membranous staining is plausible; reported cytoplasmic staining can also fit the supplied tissue observations (HPA tissue IHC). These annotations do not establish fixation sensitivity.
How strong is the staining evidence?HPA calls the tissue IHC pattern Enhanced and lists HPA007547 as IHC Enhanced, but describes only medium consistency with RNA and says presumed off-target binding was disregarded (HPA tissue IHC; HPA antibodies). Use the reported cell pattern and appropriate controls when interpreting an unexpected positive (general IHC practice).
What should IF/ICC show?HPA reports plasma membrane and cytosol localization, with ICC-IF images listed for HEK293 and SiHa; HPA071005 has ICC Supported status (HPA subcellular; HPA antibodies). This answers the localization question for IF/ICC; its staining workflow belongs in the separate IF/ICC guide.
Why is my staining missing, weak or wrong?
SituationLikely causeNext action
Late spermatids are present, but the expected High signal is absent (HPA tissue IHC).The supplied tissue reference and the observed section disagree; this alone does not identify a cause (HPA tissue IHC).Check cell identification and section quality, then review primary-antibody and detection controls and the documented IHC conditions (general IHC practice).
Signal is concentrated in nuclei rather than at cell boundaries or in cytoplasm (HPA tissue IHC).That compartment conflicts with the reported tissue pattern and cell-membrane localization (HPA tissue IHC; UniProt Q9BZM6).Treat the nuclear result as unconfirmed; compare a reported positive cell population and inspect staining controls (HPA tissue IHC; general IHC practice).
Adipocytes or HPA-negative glandular cells stain strongly (HPA tissue IHC).The pattern may reflect cross-reactivity or endogenous detection activity; HPA also notes disregarded presumed off-target binding (HPA tissue IHC; general IHC practice).Map the stained cell types and check primary-omission and detection controls before calling them ULBP1-positive (general IHC practice).
Broad, diffuse color obscures cell boundaries and the reported positive cells (HPA tissue IHC; general IHC practice).Nonspecific background or detection background can prevent a compartment-level interpretation (general IHC practice).Inspect background controls and review blocking, antibody concentration, washing and detection settings against the documented IHC method (general IHC practice).
Purkinje cells stain less strongly than late spermatids (HPA tissue IHC).That relative intensity agrees with HPA's Medium Purkinje-cell and High late-spermatid reports (HPA tissue IHC).Score each identified cell type against its own reported level; do not require equal intensity across these tissues (HPA tissue IHC; general IHC practice).
IF/ICC shows membrane or cytosolic signal that seems different from the chromogenic IHC image (HPA subcellular; HPA tissue IHC).HPA reports plasma membrane and cytosol in ICC-IF and cytoplasmic or membranous tissue IHC staining (HPA subcellular; HPA tissue IHC).Compare localization and cell identity within each application; use the separate IF/ICC guide for its controls and workflow (HPA subcellular; general IHC practice).

Sample controls for ULBP1 IHC & IF

🧪Run testis first; elongated or late spermatids should stain (HPA: High in elongated or late spermatids). Use adipose tissue as the negative comparator (HPA: adipocytes Not detected); other cells on the testis slide should provide a low-background comparison, although HPA does not establish them as ULBP1-negative (HPA: testis row specifies elongated or late spermatids only).
Positive control tissue: Testis (Elongated or late spermatids, HPA High)
Negative control tissue: Adipose tissue (HPA Not detected)
ICC-IF cell lines (HPA subcellular resource): HPA ICC-IF images show ULBP1 in HEK293, SiHa, with annotated localisation: Plasma membrane (supported), Cytosol (approved) (HPA subcellular).
Technical controls: Include no-primary (secondary-only) and species-matched rabbit IgG isotype controls, plus ULBP1-knockout tissue as a biological negative if available (selected-SKU caption: rabbit anti-ULBP1; standard IHC practice). Block endogenous peroxidase and compare testis DAB staining with the no-primary slide (selected-SKU caption: DAB detection; standard chromogenic IHC practice).
⚠️Feasibility: A target-specific fixation window and fixation effect are unreported, and the selected-SKU paraffin-section caption does not state the fixative (selected-SKU caption: fixative not stated). Retrieval dependence is unestablished, although the caption used heat-mediated EDTA at pH 8.0 (selected-SKU caption: heat-mediated EDTA retrieval). The supplied evidence does not establish whether frozen sections or IF are easier; in testis, use the counterstain and no-primary control to distinguish staining of closely packed spermatids from background DAB deposits (HPA: High in elongated or late spermatids; standard IHC practice).

Advanced ULBP1 IHC Tips

Troubleshoot ULBP1 staining in paraffin sections by checking retrieval, cell identity, subcellular pattern and controls before assigning a score.

How should I retrieve ULBP1 in paraffin sections with A04323-1?
Start with heat-mediated retrieval in EDTA at pH 8.0 (datasheet A04323-1). The selected paraffin-section image used that retrieval before incubation with 2 μg/ml primary antibody overnight at 4°C; it does not establish that retrieval works equally well after every fixation condition (datasheet A04323-1). If staining is weak, compare carefully controlled retrieval durations on serial sections while holding antibody concentration and detection constant (standard IHC practice). Check a known positive cell population and a no-primary control in each comparison, because stronger DAB alone cannot distinguish improved epitope exposure from increased background (HPA tissue IHC; standard IHC practice).
Could fixation explain weak or patchy ULBP1 staining?
Target-specific fixation sensitivity is unknown: the selected paraffin-section caption does not state a fixative (datasheet A04323-1). Record the actual fixation and processing history for each specimen, then compare sections processed under matched conditions before changing the staining protocol (standard IHC practice). Excessive or inconsistent fixation can complicate antigen detection generally, but neither the HPA staining pattern nor ULBP1 topology demonstrates a particular fixation effect for this target (standard IHC practice; HPA tissue IHC; UniProt Q9BZM6 topology). If weak staining persists, retain the established EDTA pH 8.0 retrieval as the reference condition while testing one processing variable at a time (datasheet A04323-1; standard IHC practice).
Which ULBP1 staining patterns are plausible in tissue sections?
Assess membrane-associated staining alongside cytoplasmic staining: tissue IHC reports both in testis seminiferous ducts and cerebellum (HPA tissue IHC). Plasma membrane localisation is supported and cytosol localisation is approved in cell imaging, while UniProt also records an endoplasmic-reticulum location (HPA subcellular; UniProt Q9BZM6 subcellular). ULBP1 has no transmembrane segment and is annotated as GPI-anchored, so a membrane pattern does not require a transmembrane domain (UniProt Q9BZM6 topology; UniProt Q9BZM6 keywords). In chromogenic sections, compare the apparent compartment with cell boundaries and morphology before calling diffuse pigment positive, particularly where adjacent cells overlap (standard IHC practice).
What should I check if staining changes between samples?
First confirm that sections received the same EDTA pH 8.0 retrieval and 2 μg/ml primary concentration used in the selected example (datasheet A04323-1). The record lists a signal peptide at residues 1–25, a mature chain at 26–216, and a propeptide at 217–244 (UniProt Q9BZM6 processing). It also lists one glycosylation site at residue 82 and no annotated isoforms; an epitope location for A04323-1 is not supplied (UniProt Q9BZM6 glycosylation; UniProt Q9BZM6 isoforms; datasheet A04323-1). Therefore, do not attribute staining differences to a particular isoform, cleavage event or glycan without epitope mapping or an independent assay (standard IHC practice).
How can IF help assess an ambiguous ULBP1 IHC pattern?
Use IF on a matched section to compare ULBP1 with a validated marker identifying the expected cell type, such as a macrophage marker when assessing lung macrophages (HPA tissue IHC; standard IF practice). Choose fluorophores and imaging channels after measuring tissue autofluorescence, and include single-label controls when multiplexing (standard IF practice). For an antibody recognising an extracellular, surface-accessible epitope, compare staining without permeabilisation; for an intracellular epitope, assess a separately permeabilised condition, since the A04323-1 epitope is unspecified (UniProt Q9BZM6 topology; datasheet A04323-1; standard IF practice). Treat IF as a compartment and cell-identity check, then score the chromogenic IHC sections using their own controls (standard IHC/IF practice).
How do I reduce diffuse or nonspecific DAB staining?
Check the no-primary control and inspect whether pigment follows tissue edges, necrotic areas or endogenous enzyme activity rather than intact cells (standard IHC practice). Use an appropriate peroxidase block for DAB detection, then optimise blocking and washing while keeping the selected retrieval condition at EDTA pH 8.0 (standard IHC practice; datasheet A04323-1). The selected example used 10% goat serum blocking, 2 μg/ml primary overnight at 4°C, and a biotin-based secondary system (datasheet A04323-1). Because HPA notes presumed off-target binding, require convincing cell morphology and control performance before interpreting widespread weak staining as ULBP1 (HPA tissue IHC; standard IHC practice).
How should I score ULBP1 across stained sections? ⚠ ANSWER MARKED FOR VERIFICATION
Define the compartment and cell population before scoring, since ULBP1 tissue IHC includes cytoplasmic and membranous patterns (HPA tissue IHC). For a prespecified population, report the percentage of positive intact cells and staining intensity, or combine them as an H-score using a consistent scoring rule (standard IHC practice). If counting scattered positive cells, report their density per mm² of viable tissue and normalise comparisons to the same cell population or viable area, excluding necrosis and section edges (standard IHC practice). Keep retrieval, detection, counterstain, imaging and thresholds consistent across samples, and review discordant fields against controls before aggregation (standard IHC practice).
When is ULBP1 staining credible rather than artefactual?
A credible call matches intact cell morphology and a plausible membranous or cytoplasmic compartment, with acceptable positive and no-primary controls (HPA tissue IHC; standard IHC practice). HPA reports high staining in elongated or late spermatids and medium staining in cerebellar Purkinje cells and lung macrophages, providing cell-specific reference patterns (HPA tissue IHC). Treat staining confined to section edges or necrosis, pigment in implausible compartments or cells, and signal reproduced by the no-primary control as suspect (standard IHC practice). Interpret unexpected widespread staining cautiously because HPA describes medium RNA–staining consistency and presumed off-target binding that was disregarded in its assessment (HPA tissue IHC).
