Background of IGFBP3
Insulin-like growth factor (IGF)-binding protein-3 (IGFBP-3) is a major determinant of circulating levels of the IGFs and is clinically useful for the evaluation of GH deficiency and for predicting the response to GH treatment. The circulating level of IGFBP-3 is inversely related to the risk of several common cancers, and that antiproliferative agents such as antiestrogens and retinoids act in part by up-regulating IGFBP-3 gene (IGFBP3) expression. Insulin-like growth factor-binding protein (IGFBP)-3, well characterized as the carrier of insulin-like growth factor (IGF), has been reported to have intrinsic bioactivity that is independent of IGF binding. IGFBP-3 has an IGF-independent, antiproliferative effect in undifferentiated and early differentiated but not in terminally differentiated chondrocytes. IGFBP-3 possesses both growth-inhibitory and potentiating effects on cells that are independent of IGF action and are mediated through specific. IGFBP-3 binding proteins/receptors locate at the cell membrane, cytosol, or nuclear compartments and in the extrIGFBP-3llular matrix. IGFBP3 is also located on chromosome 7. The standard product used in this kit is recombinant human IGFBP-3, consisting of 265 amino acids with the molecular mass of 29kDa. As a result of glycosylation, the molecular mass is 41KDa.

As the epitope is located somewhere on the mature form, the detection antibodies will recognize both uncleaved and cleaved forms of the protein.

A: Chelating agents such as EDTA, Heparin and Citrate can irreversibly bind metal ions from the functional domain of IGFBP-3 causing degradation of its protein structure. IGFBP-3 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes required for detection and block the antigen antibody reaction. We have tested the IGFBP-3 ELISA, treating samples with a number of anticoagulants and decided that heparin or EDTA can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

A: unable to know the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the IGFBP-3 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.

A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test IGFBP-3 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.
