Background of TNFRSF1B
Tumor necrosis factor receptor p75 (TNF-R p75, or TNF-R2) is a 75-kDa type I transmembrane protein expressed predominantly on cells of hematopoietic lineage. TNF-R p75 belongs to the TNF receptor superfamily characterized by cysteine-rich extracellular regions composed of three to six disulfide-linked domains. The tumor necrosis factor receptor II (TNFRII) gene localizes to 1p36. 2, a genomic region characteristically deleted in neuroblastomas and other malignancies. In addition, TNFRII is the principal mediator of the effects of TNF on cellular immunity, and it may cooperate with TNFRI in the killing of nonlymphoid cells. The standard product used in this kit is recombinant human sTNFRII, consisting of 24-206 amino acid sequence with the molecular mass of 20KDa.

Q: is it okay to use heparin plasma as samples in Human TNF RII Picokine® ELISA Kit (Catalog # EK0530)?

Answer
A: Chelating agents such as EDTA, Heparin and Citrate can attach metal ions from the functional domain of TNF RII causing degradation of its protein structure. TNF RII may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes required for detection and block the antigen antibody reaction. We have tested the TNF RII ELISA, treating samples with various anticoagulants and decided that heparin, EDTA or citrate can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test TNF RII for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

A: in those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer may affect immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may alter the stabilityof samples. It is recommend to assay sample on priority basis collection when possible, or aliquot into single use volumes and store samples frozen. avoid repeat freeze-thaw cycles with the stored samples to limit protein degradation.
