The IkappaB-alpha (Phospho-Tyr305) Cell-Based ELISA Kit is a convenient, lysate-free, high throughput and sensitive assay kit that can monitor IkappaB-alpha (Phospho-Tyr305) protein expression profile in cells. The kit can be used for measuring the relative amounts of IkappaB-alpha (Phospho-Tyr305) in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on IkappaB-alpha (Phospho-Tyr305).

- Microplate reader able to measure absorbance at 450 nm and/or 595 nm for Crystal Violet Cell Staining (Optional)
- Micropipettes with capability of measuring volumes ranging from 1 ul to 1 ml
- 37% formaldehyde (Sigma Cat# F-8775) or formaldehyde from other sources
- Deionized or sterile water
- Squirt bottle, manifold dispenser, multichannel pipette reservoir or automated microplate washer
- Graph paper or computer software capable of generating or displaying logarithmic functions
- Absorbent papers or vacuum aspirator
- Test tubes or microfuge tubes capable of storing 1 ml

The immunogen sequence of EKC2295 IkappaB-alpha (Phospho-Tyr305) Colorimetric Cell-Based ELISA Kit is near the N-terminus, in the general range 15 to 40aa. The definite sequence information is proprietary, thus, we cannot released it, but we can say that it is less than 25aa long.

Question
I ordered your product EKC2295 IkappaB-alpha (Phospho-Tyr305) Colorimetric Cell-Based ELISA Kit that I used for western blot and I observed beautiful band for classic form of Ikappa B alpha (37kDa). Even though I have all of this in my western blots, I have an additional band around 47kDa, and I would like to know if I could test a positive control to finally determine if it's an another form of IkappaB or not ?

Our product EKC2295 IkappaB-alpha (Phospho-Tyr305) Colorimetric Cell-Based ELISA Kit recognizes both the phosphorylated and non-phosphorylated forms of the serine 32 region of IKB alpha. It means that the band at 37kDa represents the unphosphorylated protein and the band at 47kDa is likely to be due to phosphorylation of serine residues. However, if you would like to test a positive control, we suggest to use HeLa (Human epithelial carcinoma cell line) Whole Cell Lysate.
