Background of IL-6
The human interferon-beta 2 gene (IFNB2) product is identical to that for the B-cell stimulation factor-2 (BSF-2), the hybridoma growth factor (HGF) ("interleukin-6"), and the hepatocyte stimulating factor (HSF). Proteins derived from this gene mediate the plasma protein response to tissue injury (acute-phase response) and regulate the growth and differentiation of both B and T cells. Interleukin-6 (IL6) has come to be regarded as a potential osteoporotic factor because it has stimulatory effects on cells of the osteoclast lineage, and, thus, may play a role in the pathogenesis of bone loss associated with estrogen deficiency. IL-6 has many roles essential to the regulation of the immune response, hematopoiesis, and bone resorption. It is involved not only in the hepatic acute phase response but also in adipose tissue metabolism, lipoprotein lipase activity, and hepatic triglyceride secretion. Overproduction of IL-6, a proinflammatory cytokine, is associated with a spectrum of age-related conditions including cardiovascular disease, osteoporosis, arthritis, type 2 diabetes, certain cancers, periodontal disease, frailty, and functional decline. BSF-2 is a novel interleukin consisting of 184 amino acids. The standard product used in this kit is recombinant IL-6, consisting of 188 amino acids with the molecular mass of 21.8kDa.

"The kit contained an extra standard which is very useful if you need to optimize your dilution factor before you use the rest of your samples. The instructions were very easy to follow and the results we obtained were consistent between replicates. Very easy to use. Don't waste your time or money [on other kits]; just use this kit." (Customer’s Tip: "Optimize dilutions 1:5-1:250.")

IL-6 Estimation in Cell Culture Supernatant--Divya Sharma, NDSU, Pharmaceutical Sciences, Graduate Student
Excellent

"Good estimation of IL-6. The results were comparable and reproducible each time. Good brand name, very good quality of reagents. Overall justified results with quality of the product. Also good value for the money. Order 4 or more kits to get good discount. That was helpful in my case. Will definitely purchase again throughout my project." (Customer’s Tip: "If performing for 6-well plate supernatant, may run a little higher in concentration. Consider diluting samples on doing a test run.")

Question
Q: which procedure should I follow in order to thaw whole blood sample for IL-6 ELISA after freezing?
G. Maharaj
Verified customer
Asked: 2020-03-14
Answer
A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test IL-6 for a later time, you should let the blood clot in glass tubes and separate the serum to freeze for later analysis.

Question
What standard curve (Linear, 4PL or Hyperbolic) is recommended to use in finding out the coresponding concentrations in our data using EK0411 kit?
Verified customer
Asked: 2020-01-08
Answer
For Mouse IL-6 ELISA Kit PicoKine EK0411, if your sample data points are within the middle of the curve, i.e. in the near linear region of the curve, then you can use the best fitting curve out of hyperbola, 4PL and 5PL. However, if your sample data points are near the ends of the standard curve, where it becomes not linear, it is recommended that you use 4PL for fitting, as it best fits the nature of the assay. It is also recommended that you adjust your assay so that the sample data points fall into the middle linear range of the curve. For further troubleshooting you may send us your generated data.

Question
Q: for how much duration can samples (cell cultures, serum, and plasma) be stored and still be stable for analysis IL-6 using the EK0411 Mouse IL-6 Picokine® ELISA Kit?
Verified Customer
Verified customer
Asked: 2019-12-20
Answer
A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may alter the stabilityof samples. It is recommend to assay sample immediately after collection when possible, or aliquot into single use volumes and store samples frozen. avoid repetitive freeze-thaw cycles with the stored samples to prevent protein degradation.

Question
Q: can you tell me how to prepare cell lysates prepared for use in Picokine® ELISA kits?
Verified Customer
Verified customer
Asked: 2018-12-11
Answer
A: for those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer may impact immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

Question
Q: Can IL-6 ELISA Kits be used with tissue homogenates (or other non-validated sample types)?
Verified Customer
Verified customer
Asked: 2018-12-10
Answer
A: Unfortunately, Boster Bio has not routinely validated tissue homogenates as a sample type for ELISA kits. This does not mean that ELISA kits are not suitable for other sample types than we have tested: it means further investigation is needed. One will need to perform a spike and recovery study to determine if an unvalidated sample type will work with a particular kit. To perform a spike and recovery experiment, one should divide a sample into two aliquots. In one of the aliquots, the user should spike in a known amount of the kit standard. a dilution series is performed comparing the spiked versus the unspiked sample. Generally, samples with expected recovery and linearity between 80-120% are considered acceptable. This method can be used to validate any sample type that has not been assessd by Boster Bio. for a more detailed spike and recovery protocol, please contact technical support.
Note: acceptable ranges should be determined individually by each laboratory. Additionally, technical support can help determine if a buffer component is not compatible with a given ELISA kit. please see the Citations tab on the product webpage for peer-reviewed papers utilizing a wide range of sample types. We also have an innovator's reward program where if the user validates our ELISA kits in applications or samples previously not validated by Boster Bio or other users, and share such information with us by submit a review, we will reward the user's efforts with a free antibody or ELISA kit from our catalog. Biocompare.com will also give $20 Amazon giftcard as an additional reward, if the review is submitted there as well.

Question
Q: Are Boster Bio recombinant proteins and antibodies sterile?
Verified Customer
Verified customer
Asked: 2015-10-08
Answer
A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

Question
Q: if the enzyme conjugated IL-6 antibodies are mixed with the substrate, will that convert the substrate into the enzymatic reaction product? Or the enzyme function is only activated when the antibody is attached with the IL-6 antigen?
Verified Customer
Verified customer
Asked: 2015-02-09
Answer
A: The enzyme is always active. Avoid contaminating the substrate with enzyme prior to the incubation otherwise it compromises the assay with false positive signal.
