Background of IL6
The human interferon-beta 2 gene (IFNB2) product is identical to that for the B-cell stimulation factor-2 (BSF-2), the hybridoma growth factor (HGF) ("interleukin-6"), and the hepatocyte stimulating factor (HSF). Proteins derived from this gene mediate the plasma protein response to tissue injury (acute-phase response) and regulate the growth and differentiation of both B and T cells. Interleukin-6 (IL6) has come to be regarded as a potential osteoporotic factor because it has stimulatory effects on cells of the osteoclast lineage, and, thus, may play a role in the pathogenesis of bone loss associated with estrogen deficiency. IL-6 has many roles essential to the regulation of the immune response, hematopoiesis, and bone resorption. It is involved not only in the hepatic acute phase response but also in adipose tissue metabolism, lipoprotein lipase activity, and hepatic triglyceride secretion. Overproduction of IL-6, a proinflammatory cytokine, is associated with a spectrum of age-related conditions including cardiovascular disease, osteoporosis, arthritis, type 2 diabetes, certain cancers, periodontal disease, frailty, and functional decline. BSF-2 is a novel interleukin consisting of 184 amino acids. The standard product used in this kit is recombinant IL-6, consisting of 188 amino acids with the molecular mass of 21.8kDa.

A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test IL-6 for a later time, you should let the blood clot in glass tubes and separate the serum to freeze for later analysis.

For Mouse IL-6 ELISA Kit PicoKine EK0411, if your sample data points are within the middle of the curve, i.e. in the near linear region of the curve, then you can use the best fitting curve out of hyperbola, 4PL and 5PL. However, if your sample data points are near the ends of the standard curve, where it becomes not linear, it is recommended that you use 4PL for fitting, as it best fits the nature of the assay. It is also recommended that you adjust your assay so that the sample data points fall into the middle linear range of the curve. For further troubleshooting you may send us your generated data.

A: Unfortunately, Boster Bio has not routinely validated tissue homogenates as a sample type for ELISA kits. This does not mean that ELISA kits are not suitable for other sample types than we have tested: it means further investigation is needed. One will need to perform a spike and recovery study to determine if an unvalidated sample type will work with a particular kit. To perform a spike and recovery experiment, one should divide a sample into two aliquots. In one of the aliquots, the user should spike in a known amount of the kit standard. a dilution series is performed comparing the spiked versus the unspiked sample. Generally, samples with expected recovery and linearity between 80-120% are considered acceptable. This method can be used to validate any sample type that has not been assessd by Boster Bio. for a more detailed spike and recovery protocol, please contact technical support.
Note: acceptable ranges should be determined individually by each laboratory. Additionally, technical support can help determine if a buffer component is not compatible with a given ELISA kit. please see the Citations tab on the product webpage for peer-reviewed papers utilizing a wide range of sample types.
