Materials Required But Not Provided
- Microplate reader in standard size.
- Automated plate washer.
- Adjustable pipettes and pipette tips. Multichannel pipettes are recommended in the condition of large amount of samples in the detection.
- Clean tubes and Eppendorf tubes.
- Washing buffer (neutral PBS or TBS).
- Preparation of 0.01M TBS: Add 1.2g Tris, 8.5g NaCl; 450μl of purified acetic acid or 700μl of concentrated hydrochloric acid to 1000ml H2
- Preparation of 0.01 M PBS: Add 8.5g sodium chloride, 1.4g Na O and adjust pH to 7.2-7.
Typical Data Obtained from Mouse WISP1/CCN4 PicoKine™ ELISA Kit
(TMB reaction incubate at 37°C for 25-30min)
Intra/Inter Assay Precision
|Intra-Assay Precision||Inter-Assay Precision|
Three samples with differing target protein concentrations were assayed using four different lots to measure the CV% lot to lot variance.
To assay reproducibility, three samples with differing target protein concentrations were assayed using four different lots.
|Lots||Lot1 (pg/ml)||Lot2 (pg/ml)||Lot3 (pg/ml)||Lot4 (pg/ml)||Mean (pg/ml)||Standard Deviation||CV (%)|
*The typical data is obtained from Boster's internal QC result and for reference only. It may differ from the lab test results of the end users. It is more important that the user's lab test results reflect the same linearity demonstrated in the typical data than achieving exactly the same O.D. values.

|Tissue Specificity||Highly expressed in kidney and lung. Lower levels in heart, brain, spleen, liver, skeletal muscle and testis. Expressed in low metastatic melanoma cells.|
|Alternative Names||WNT1-inducible-signaling pathway protein 1;WISP-1;CCN family member 4;ELM-1;Wisp1;Ccn4, Elm1;|
|Subcellular Localization||Secreted .|
|Molecular Weight||40703 MW|
*if product is indicated to react with multiple species, protein info is based on the human gene.
|Protein Function||Downstream regulator in the Wnt/Frizzled-signaling pathway (By similarity). Associated with cell survival. Adheres to skin and melanoma fibroblasts (By similarity). In vitro binding to skin fibroblasts occurs through the proteoglycans, decorin and biglycan (By similarity). Suppresses tumor growth in vivo. .|
|Background||WISP1(WNT1-Inducible Signaling Pathway Protein 1), also known as CCN4, is a matricellular protein that in humans is encoded by the WISP1 gene. WISP1 is induced by WNT1 and belongs to the CCN family, which includes connective tissue growth factor, cysteine-rich-61, and nephroblastoma overexpressed. By use of radiation hybrid mapping panels, the WISP1 was mapped to chromosome 8q24.1-q24.3, roughly 4 Mb distal to MYC. It found that 2 distinct systems demonstrated that WISP induction was associated with expression of WNT1. WISP1 genomic DNA was amplified in colon cancer cell lines and in human colon tumors, and its RNA was overexpressed in 84% of tumors examined compared with patient-matched normal mucosa.|
