Electrophoresis was performed on a 7.5% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: normal drosophila melanogaster head tissues,
Lane 2: CG2727 KO / +,
Lane 3: normal drosophila melanogaster thorax tissues,
Lane 4: CG2727 KO / +.
After electrophoresis, proteins were transferred to PVDF at 20 V overnight. Blocked the membrane with 5% skim milk in PBS-T for 30 min at RT. The membrane was incubated with rabbit anti-Emp antigen affinity purified polyclonal antibody (Catalog # DZ41152) at 1:1000 dilution in PBS-T with 1% skim milk for 1h at RT, then washed with PBS 3 times and probed with a goat anti-rabbit IgG-HRP secondary antibody at 1:10000 dilution PBS-T with 1% skim milk for 1h at RT, then washed with PBS 3 times. The signal is detected with Immobilon Forte Western HRP substrate. A specific band was detected for Emp at approximately 59 kDa.
