Description
Serotonin signaling (putative): HTR2B (Gq/PLCβ→IP₃/Ca²⁺, MAPK) in vascular/valvular biology and fibrosis research. Assay: rabbit polyclonal for WB (human/rat). Can be analyzed alongside neutrophil protease activity via Elastase ELISA and anti-angiogenic axis sVEGFR1/sFLT1 ELISA to relate GPCR signaling to vascular remodeling (putative).
Storage & Handling
Store at -20˚C for one year from date of receipt. After reconstitution, at 4˚C for one month. It can also be aliquotted and stored frozen at -20˚C for six months. Avoid repeated freeze-thaw cycles.

Background of HTR2B
5HT2B Receptor is known as HTR2B. This gene encodes one of the several different receptors for 5-hydroxytryptamine (serotonin) that belongs to the G-protein coupled receptor 1 family. Serotonin is a biogenic hormone that functions as a neurotransmitter, a hormone, and a mitogen. Serotonin receptors mediate many of the central and peripheral physiologic functions of serotonin, including regulation of cardiovascular functions and impulsive behavior. Population and family-based analyses of a minor allele (glutamine-to-stop substitution, designated Q20*) which blocks expression of this protein, and knockout studies in mice, suggest a role for this gene in impulsivity. However, other factors, such as elevated testosterone levels, may also be involved.

Western blot analysis of 5HT2B Receptor using anti-5HT2B Receptor antibody (PB9600).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Brain Tissue Lysate at 50ug,
Lane 2: U87 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-5HT2B Receptor antigen affinity purified polyclonal antibody (Catalog # PB9600) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for 5HT2B Receptor at approximately 54 kDa. The expected band size for 5HT2B Receptor is at 54 kDa.

We are currently using anti-5HT2B Receptor/HTR2B antibody PB9600 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on dog tissues as well?

The anti-5HT2B Receptor/HTR2B antibody (PB9600) has not been tested for cross reactivity specifically with dog tissues, but there is a good chance of cross reactivity.
