Background of ACTA1
Actin, a highly conserved protein, is a major component of both the cytoskeletal and contractile structures in the cell types. It varies in amount, being related to the type of differentiation and to the functional state of cells and tissues. The actins exhibit over 90% sequence homology, but each isoform has a unique NH2-terminal sequence. The isoforms are comprised of three alpha-actin, one beta-actin, two gamma-actin. Because the amino acid sequence of the C-terminal is the same for almost all actins, this antibody has been raised using a synthetic peptide corresponding to the C-terminal 11 residues.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Spleen Tissue Lysate at 50ug,
Lane 2: Mouse Spleen Tissue Lysate at 50ug,
Lane 3: HELA Whole Cell Lysate at 40ug,
Lane 4: MCF-7 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Actin antigen affinity purified polyclonal antibody (Catalog # PB9751) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Actin at approximately 42 kDa. The expected band size for Actin is at 42 kDa.

Actin was detected in a paraffin-embedded section of mouse spleen tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 μg/ml rabbit anti-Actin Antibody (PB9751) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.

My boss were content with the WB result of your anti-Actin/ACTA1 antibody. However we have been able to see positive staining in biceps brachii cytoskeleton. using this antibody. Is that expected? Could you tell me where is ACTA1 supposed to be expressed?
