Flow Cytometry analysis of 293T cells using anti-AFF4 antibody (M03824).
Overlay histogram showing 293T cells stained with M03824 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with mouse anti- AFF4 Antibody (M03824, 1μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-mouse IgG (BA1126, 5-10μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was mouse IgG (1μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

Background of AFF4
The AFF4 gene encodes a scaffold protein that functions as a core component of the super elongation complex (SEC), which is involved in transcriptional regulation during embryogenesis. The protein encoded by this gene belongs to the AF4 family of transcription factors involved in leukemia. It is a component of the positive transcription elongation factor b (P-TEFb) complex. This gene is mapped to chromosome 5q31.
