Background of Aquaporin-4

Aquaporin 4 is found in the basolateral cell membrane of principal collecting duct cells and provides a pathway for water to exit these cells. The gene of AQP4 is mapped to 18q11.2-q12.1. Similar to other aquaporins, the AQP4 gene is composed of 4 exons encoding 127, 55, 27, and 92 amino acids separated by introns of 0.8, 0.3, and 5.2 kb. Unlike other aquaporins, an alternative coding initiation sequence (designated exon 0) was located 2.7 kb upstream of exon 1. When spliced together, M1 and the subsequent 10 amino acids are encoded by exon 0; the next 11 amino acids and M23 are encoded by exon 1. AQP4 is expressed in astrocytes and is upregulated by direct insult to the central nervous system. And AQP4 is the predominant water channel in the brain and has an important role in brain water homeostasis. It is abundant in mammalian brain and is concentrated in astrocytic foot processes at the blood-brain barrier.

Antibody Validation

Question

Would anti-Aquaporin 4/AQP4 antibody PB9475 work on primate IHC with lateral globus pallidus?

Verified Customer

Verified customer

Asked: 2020-02-20

Answer

Our lab technicians have not validated anti-Aquaporin 4/AQP4 antibody PB9475 on primate. You can run a BLAST between primate and the immunogen sequence of anti-Aquaporin 4/AQP4 antibody PB9475 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated primate samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in primate lateral globus pallidus in IHC, you can get your next antibody for free.

Question

Would PB9475 anti-Aquaporin 4/AQP4 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2020-01-16

Answer

It shows on the product datasheet, PB9475 anti-Aquaporin 4/AQP4 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

My question regarding product PB9475, anti-Aquaporin 4/AQP4 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

M. Krishna

Verified customer

Asked: 2013-01-23

Answer

We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free PB9475 anti-Aquaporin 4/AQP4 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
