Flow Cytometry analysis of Jurkat cells using anti-c-Myb antibody (PB9288).
Overlay histogram showing Jurkat cells stained with PB9288 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-c-Myb Antibody (PB9288, 1 μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10 μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1 μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

Background of MYB
Myb proto-oncogene protein, also known as transcriptional activator Myb, is a protein that in humans is encoded by the MYB gene. It is a member of the MYB (myeloblastosis) family of transcription factors. This gene is mapped to 6q23.3.The protein contains three domains, an N-terminal DNA-binding domain, a central transcriptional activation domain and a C-terminal domain involved in transcriptional repression. This protein plays an essential role in the regulation of hematopoiesis and may play a role in tumorigenesis, including the regulation of miR-155 in B-cells. MYB is also a critical target of MIR150.
