Background of Carbonic Anhydrase XII/CA12
Carbonic anhydrase 12 is an enzyme that in humans is encoded by the CA12 gene. It is mapped to 15q22.2. Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva, and gastric acid. This gene product is a type I membrane protein that is highly expressed in normal tissues, such as kidney, colon and pancreas, and has been found to be overexpressed in 10% of clear cell renal carcinomas. Three transcript variants encoding different isoforms have been identified for this gene.
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence and ELISA with known positive and negative samples to ensure specificity and high affinity.

Figure 1. Western blot analysis of CA12 using anti-CA12 antibody (A04063-1).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human U2OS whole cell lysates,
Lane 2: human Hela whole cell lysates,
Lane 3: human T-47D whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CA12 antigen affinity purified polyclonal antibody (Catalog # A04063-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CA12 at approximately 49KD. The expected band size for CA12 is at 39KD.
