Background of CA3
Carbonic anhydrase III (CA3) is an enzyme that in humans is encoded by the CA3 gene. CA3 is a member of a multigene family (at least six separate genes are known) that encode carbonic anhydrase isozymes. The gene spans 10.3 kb and contains seven exons and six introns. Using a cDNA clone of the CA3 gene in the study of human-rodent hybrids, the gene was mapped to chromosome 8 which carries a cluster of CA genes. CA3 is found in high concentration in muscle. CA3 is used in conjunction with creatine kinase detection of the carrier state for Duchenne muscular dystrophy.

Western blot analysis of CA3 using anti-CA3 antibody (PA1439).
Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human THP-1 whole cell lysates,
Lane 2: rat liver tissue lysates,
Lane 3: rat fat tissue lysates,
Lane 4: mouse liver tissue lysates,
Lane 5: mouse fat tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CA3 antigen affinity purified polyclonal antibody (PA1439) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for CA3 at approximately 30 kDa. The expected band size for CA3 is at 30 kDa.
