Background of Tnfrsf4
Tumor necrosis factor receptor superfamily, member 4, also known as ACT35 or CD134, is a cell surface glycoprotein that was discovered through the production of a monoclonal antibody raised against the HUT-102 cell line. It belongs to the tumor necrosis factor receptor superfamily. CD134 was mapped to 1p36 by fluorescence in situ hybridization. CD134 is the primary receptor for feline immunodeficiency virus. And CD134 expression can promote viral binding and renders cells permissive for viral entry, productive infection, and syncytium formation. Stimulating the receptor can improve the response to a powerful virus vector and may be useful in vaccine development.

Western blot analysis of CD134/OX40 using anti-CD134/OX40 antibody (A02495).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat heart tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD134/OX40 antigen affinity purified polyclonal antibody (Catalog # A02495) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CD134/OX40 at approximately 45KD. The expected band size for CD134/OX40 is at 29KD.
