Background of CD244

CD244 (Cluster of Differentiation 244) is a human protein encoded by the CD244 gene. It is also known as Natural Killer Cell Receptor 2B4. Tangye et al. (1999) mapped the 2B4 gene to 1q22. Suzuki et al. (2008) identified a functional single-nucleotide polymorphism (SNP) in the CD244 gene that contributes to rheumatoid arthritis susceptibility. Functional analysis by Boles et al. (1999) demonstrated that engagement of 2B4 with specific antibody activates NK cytolytic activity. Using recombinant human NK cell-activating ligand 2B4 fused to domains 3 and 4 of rodent Cd4 and flow cytometric analysis, Brown et al. (1998) demonstrated that CD48 binds to 2B4. Watzl et al. (2000) showed that antibody-mediated cross-linking of 2B4 leads to its rapid tyrosine phosphorylation, which is necessary for 2B4-mediated killer cell activity.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

4 Customer Q&As for Anti-CD244 Antibody Picoband®

Question

Does PA2057 anti-CD244 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Asked: 2020-01-10

Answer

As indicated on the product datasheet, PA2057 anti-CD244 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

Does anti-CD244 antibody PA2057 work on monkey WB with natural killer cell?

Verified Customer

Verified customer

Asked: 2019-09-06

Answer

Our lab technicians have not validated anti-CD244 antibody PA2057 on monkey. You can run a BLAST between monkey and the immunogen sequence of anti-CD244 antibody PA2057 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated monkey samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in monkey natural killer cell in WB, you can get your next antibody for free.

Question

We are currently using anti-CD244 antibody PA2057 for human tissue, and we are well pleased with the IHC results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on dog tissues as well?

M. Jones

Verified customer

Asked: 2016-07-06

Answer

The anti-CD244 antibody (PA2057) has not been tested for cross reactivity specifically with dog tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in dog you can get your next antibody for free. Please contact me if I can help you with anything.
