Flow Cytometry analysis of 293T cells using anti-CYP11B1/C11B2/CYP11B2 antibody (A03766-3).
Overlay histogram showing 293T cells stained with A03766-3 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-CYP11B1/C11B2/CYP11B2 Antibody (A03766-3,1μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

Background of CYP11B1/2
CYP11B1 shows close homology to the CYP11B2 gene, which encodes aldosterone synthase and is normally expressed only in the zona glomerulosa. Both genes map to chromosome 8q21. This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the mitochondrial inner membrane.
