Background of DKK2
Dickkopf-related protein 2 is a protein that in humans is encoded by the DKK2 gene. This gene encodes a protein that is a member of the dickkopf family. The secreted protein contains two cysteine rich regions and is involved in embryonic development through its interactions with the Wnt signaling pathway. It can act as either an agonist or antagonist of Wnt/beta-catenin signaling, depending on the cellular context and the presence of the co-factor kremen 2. Activity of this protein is also modulated by binding to the Wnt co-receptor LDL-receptor related protein 6 (LRP6).

Western blot analysis of DKK2 using anti-DKK2 antibody (PB9551).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Cardiac Muscle Tissue Lysate at 50ug,
Lane 2: Rat Brain Tissue Lysate at 50ug,
Lane 3: A549 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-DKK2 antigen affinity purified polyclonal antibody (Catalog # PB9551) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for DKK2 at approximately 28 kDa. The expected band size for DKK2 is at 28 kDa.

We are currently using anti-DKK2 antibody PB9551 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, rat. Is it likely that the antibody can work on dog tissues as well?

The anti-DKK2 antibody (PB9551) has not been tested for cross reactivity specifically with dog tissues, though there is a good chance of cross reactivity.
