Background of EBAG9
Receptor-binding cancer antigen expressed on SiSo cells is a protein that in humans is encoded by the EBAG9 gene. This gene was identified as an estrogen-responsive gene. Regulation of transcription by estrogen is mediated by estrogen receptor, which binds to the estrogen-responsive element found in the 5'-flanking region of this gene. And the encoded protein is a tumor-associated antigen that is expressed at high frequency in a variety of cancers. Alternate splicing results in multiple transcript variants. A pseudogene of this gene has been defined on chromosome 10.

Western blot analysis of EBAG9 using anti-EBAG9 antibody (PB9553).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Testis Tissue Lysate at 50ug,
Lane 2: 22RV1 Whole Cell Lysate at 40ug,
Lane 3: HELA Whole Cell Lysate at 40ug,
Lane 4: MCF-7 Whole Cell Lysate at 40ug,
Lane 5: JURKAT Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-EBAG9 antigen affinity purified polyclonal antibody (Catalog # PB9553) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for EBAG9 at approximately 34 kDa. The expected band size for EBAG9 is at 34 kDa.

EBAG9 was detected in a paraffin-embedded section of mouse testis tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 μg/ml rabbit anti-EBAG9 Antibody (PB9553) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.

Question
We are currently using anti-EBAG9 antibody PB9553 for human tissue, and we are happy with the IHC results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on horse tissues as well?

The anti-EBAG9 antibody (PB9553) has not been validated for cross reactivity specifically with horse tissues, but there is a good chance of cross reactivity.
