Background of Cfd
Factor D is a protein which in humans is encoded by the CFD gene. The protein encoded by this gene is a member of the trypsin family of peptidases. The encoded protein is a component of the alternative complement pathway best known for its role in humoral suppression of infectious agents. It is also a serine protease that is secreted by adipocytes into the bloodstream. And it stimulates glucose transport for triglyceride accumulation in fats cells and inhibits lipolysis. Finally, the encoded protein has a high level of expression in fat, suggesting a role for adipose tissue in immune system biology.

Western blot analysis of Factor D using anti-Factor D antibody (PB9850).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: mouse spleen tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Factor D antigen affinity purified polyclonal antibody (Catalog # PB9850) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Factor D at approximately 43 kDa. The expected band size for Factor D is at 28 kDa.

We are currently using anti-Factor D/CFD antibody PB9850 for mouse tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says mouse. Is it possible that the antibody can work on canine tissues as well?

The anti-Factor D/CFD antibody (PB9850) has not been validated for cross reactivity specifically with canine tissues, but there is a good chance of cross reactivity.
