Background of GABRB3
This gene encodes a member of the ligand-gated ionic channel family. The encoded protein is one the subunits of a multi-subunit chloride channel that serves as the receptor for gamma-aminobutyric acid, a major inhibitory neurotransmitter of the mammalian nervous system. And this gene is located on the long arm of chromosome 15 in a cluster with two other genes encoding related subunits of the family. It may be associated with the pathogenesis of several disorders including Angelman syndrome, Prader-Willi syndrome, nonsyndromic orofacial clefts, epilepsy and autism. Alternatively spliced transcript variants encoding distinct isoforms have been described.

Western blot analysis of GABRB3 using anti-GABRB3 antibody (PB9595).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Brain Tissue Lysate at 50ug,
Lane 2: Mouse Brain Tissue Lysate at 50ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GABRB3 antigen affinity purified polyclonal antibody (Catalog # PB9595) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for GABRB3 at approximately 54 kDa. The expected band size for GABRB3 is at 54 kDa.

We are currently using anti-GABA A Receptor beta 3/GABRB3 antibody PB9595 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on horse tissues as well?

The anti-GABA A Receptor beta 3/GABRB3 antibody (PB9595) has not been tested for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity.
