Background of Golgi Glycoprotein 1/GLG1

This monoclonal antibody recognizes a protein of 134kDa, which binds fibroblast growth factor and E-selectin (cell-adhesion lectin on endothelial cells mediating the binding of neutrophils). Fucosylation is essential for binding to E-selectin. It contains sialic acid residues and 16 Cys-rich GLG1 repeats. This monoclonal antibody can be used to stain the Golgi complex in cell or tissue preparations and can be used as a Golgi marker in subcellular fractions. It produces a diffuse staining pattern of the Golgi zone in normal and malignant cells. This monoclonal antibody is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells. The Golgi apparatus is an organelle present in all eukaryotic cells that forms a part of the endomembrane system. The primary function of the Golgi apparatus is to process and package macromolecules synthesized by the cell for exocytosis or use within the cell. The Golgi is made up of a stack of flattened, membrane-bound sacs known as cisternae, with three functional regions: the cis face, medial region and trans face. Each region consists of various enzymes that selectively modify the macromolecules passing though them, depending on where they are destined to reside. Several spherical vesicles that have budded off of the Golgi are present surrounding the main cisternae. The Golgi tends to be more pronounced and numerous in cells that make and secrete many substances such as plasma B cells.

Antibody Validation

Assay Dilutions Recommendation

The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.

Immunohistochemistry (Formalin-fixed) (0.1-0.2ug/ml for 30 min at RT)(Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 minutes)
Optimal dilution for a specific application should be determined.

Question

Would M07510 anti-GLG1 (Golgi Glycoprotein 1) (Marker for Human Cells) Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2017-08-04

Answer

As indicated on the product datasheet, M07510 anti-GLG1 (Golgi Glycoprotein 1) (Marker for Human Cells) Monoclonal antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

Would anti-GLG1 (Golgi Glycoprotein 1) (Marker for Human Cells) Monoclonal antibody M07510 work on primate IHC with testis?

S. Mitchell

Verified customer

Asked: 2016-12-27

Answer

Our lab technicians have not validated anti-GLG1 (Golgi Glycoprotein 1) (Marker for Human Cells) Monoclonal antibody M07510 on primate. You can run a BLAST between primate and the immunogen sequence of anti-GLG1 (Golgi Glycoprotein 1) (Marker for Human Cells) Monoclonal antibody M07510 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated primate samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in primate testis in IHC, you can get your next antibody for free.
