Background of Glycophorin A
Recognizes a sialoglycoprotein of 39kDa, identified as glycophorin A (GPA). It is present on red blood cells (RBC) and erythroid precursor cells. It has been shown that glycophorin acts as the receptor for Sandei virus and parvovirus. Glycophorins A (GPA) and B (GPB), which are single, trans-membrane sialoglycoproteins. GPA is the carrier of blood group M and N specificities, while GPB accounts for S and U specificities. GPA and GPB provide the cells with a large mucin like surface and it has been suggested this provides a barrier to cell fusion, so minimizing aggregation between red blood cells in the circulation.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Assay Dilutions Recommendation
The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.
Western Blot (1-2ug/ml)
Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 minutes at RT)(Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 minutes)
Optimal dilution for a specific application should be determined.

Asked: 2020-03-17
Answer
Our lab technicians have not tested anti-Glycophorin A / CD235a (Erythrocyte Marker) Monoclonal antibody M02184 on bovine. You can run a BLAST between bovine and the immunogen sequence of anti-Glycophorin A / CD235a (Erythrocyte Marker) Monoclonal antibody M02184 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated bovine samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in bovine blood in IHC, you can get your next antibody for free.

Question
Will M02184 anti-Glycophorin A / CD235a (Erythrocyte Marker) Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
C. Krishna
Verified customer
Asked: 2016-10-12
Answer
As indicated on the product datasheet, M02184 anti-Glycophorin A / CD235a (Erythrocyte Marker) Monoclonal antibody as been validated on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
