Background of CXCL3
Chemokine (C-X-C motif) ligand 3 (CXCL3), also known as GRO protein gamma (GROg) and macrophage inflammatory protein-2-beta (MIP2b), is a small cytokine belonging to the CXC chemokine family. It is mapped to 14p2. CXCL3 controls migration and adhesion of monocytes and mediates its effects on its target cell by interacting with a cell surface chemokine receptor. It has been shown that CXCL3 regulates cell autonomously the migration of the precursors of cerebellar granule neurons toward the internal layers of cerebellum, during the morphogenesis of cerebellum. CXCL3 also play fundamental roles in the development, homeostasis and it has effects on cells of the central nervous system as well as on endothelial cells involved in angiogenesis or angiostasis.

Western blot analysis of GRO gamma using anti-GRO gamma antibody (PB9941).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: mouse lung tissue lysates,
Lane 2: mouse spleen tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GRO gamma antigen affinity purified polyclonal antibody (Catalog # PB9941) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for GRO gamma at approximately 25 kDa. The expected band size for GRO gamma is at 11 kDa.

We are currently using anti-GRO gamma/CXCL3 antibody PB9941 for mouse tissue, and we are happy with the ELISA results. The species of reactivity given in the datasheet says mouse. Is it possible that the antibody can work on feline tissues as well?
