Background of HNF1B
HNF1 homeobox B (hepatocyte nuclear factor 1 homeobox B), also known as HNF1B or transcription factor 2 (TCF2), is a human gene. It is a member of the homeodomain-containing superfamily of transcription factors. This gene is mapped to 17q12. The HNF1B protein is believed to form heterodimers with another liver-specific member of this transcription factor family, TCF1. HNF1B functions as both a classic transcriptional activator and as a bookmarking factor that marks target genes for rapid transcriptional reactivation after mitosis. HNF1B also can regulate renal tubulogenesis by controlling expression of SOC3. Mutation of HNF1B that disrupts normal function has been identified as the cause of MODY5 (Maturity-Onset of Diabetes, Type 5).

Western blot analysis of HNF1 beta using anti-HNF1 beta antibody (PB9597).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Liver Tissue Lysate at 50ug,
Lane 2: Rat Kidney Tissue Lysate at 50ug,
Lane 3: SW620 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-HNF1 beta antigen affinity purified polyclonal antibody (Catalog # PB9597) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for HNF1 beta at approximately 61 kDa. The expected band size for HNF1 beta is at 61 kDa.

We are currently using anti-HNF1 beta/HNF1B antibody PB9597 for human tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, rat. Is it likely that the antibody can work on monkey tissues as well?

The anti-HNF1 beta/HNF1B antibody (PB9597) has not been validated for cross reactivity specifically with monkey tissues, though there is a good chance of cross reactivity.
